Superiority

Rifampicin is an antibiotic that potently inhibits RNA polymerase and blocks RNA synthesis in Escherichia coli. Rifampicin has been shown to inhibit α-synuclein fibrillation and disaggregate fibrils in a concentration-dependent manner. Data indicates that Rifampicin can activate the nuclear pregnane X receptor, which affects cytochrome P450, and the activity of glucuronosyltransferases and P-glycoprotein. Rifampicin has been shown to enhance CYP2C-mediated metabolism, affect compounds that are transported by P-glycoprotein and metabolized by CYP3A4. Rifampicin has been used in tuberculosis studies.

Rifampicin is a rifamycin antibiotic and inhibitor of bacterial RNA polymerase (IC50 = 0.01 μg/ml for the E. coli enzyme). It inhibits the growth of M. tuberculosis H37Rv in mouse peritoneal macrophages (MIC = 0.8 μg/ml) as well as clinical isolates of various species of StaphylococcusStreptococcusHaemophilus, and Neisseria (MICs = 0.009-1.4 μg/ml). Rifampicin increases survival in a mouse model of tuberculosis infection. It is also an agonist of the human pregnane X receptor (PXR; EC50 = ~2 μM). Formulations containing rifampicin have been used in the treatment of tuberculosis and meningococcal carriers.

Details

Antibiotic that specifically inhibits DNA-dependent RNA polymerase in bacteria by forming an inactive complex. Does not affect mammalian RNA polymerase. Inhibits transcription by preventing the initial transcription complex from entering the elongation mode.

Antibiotic effective against mycobacteria. Can also inhibit DNA polymeraseRifampin is an antibiotic used in the treatment of tuberculosis and its related microbacterial infections. It is involved in the inhibition of DNA polymerase and RNA polymerase. As an antipruritic agent, it is used in the autoimmune cholestatic liver disease and primary biliary cirrhosis (PBC).

Rifampicin (200, 400 mg/kg) can induce fatty liver at high concentration. Rifampicin (30 mg/kg, i.p.) treatment of S464P biofilms in vivo results in a slight decline, but earlier rebinds in bioluminescence from these catheters compared with the parental signal, whereas rifampicin has no affect on bioluminescence in mice infected with mutant H481Y.
